Hello!
As usual, I am here with a small problem.
A drug company has recently come up with 'omeprazole coated with alkali'. As far as my knowledge of pharmacology is concerned, I think any antacid (alkali, H2 blocker...etc.) is contraindicated with Omeprazole (and other PPI) because it ionizes ionto sulphenic and sulphanillic (..or something like that) ions at pH <4; which can not be attained if other antacids are used.
Now the questions:
1. Do PPI block Na-K ATPase in oxyntic cells from luminal side (i.e locally, from the stomach) or from interstitial (i.e reach the cells by blood) side?
2. If they act from the interstitial side, where do they form the charged ions?
3. What is the site of absorption of PPI?
4. Is the new combination rational?
